What Is Triprolidine and How Does It Work?

Triprolidine is an over-the-counter medication used for the treatment of allergic rhinitis.

What Are Side Effects Associated with Using Triprolidine?

Common side effects of Triprolidine include:

  • drowsiness, 
  • dizziness, 
  • headache, 
  • nausea, 
  • vomiting, 
  • loss of appetite, 
  • constipation, 
  • stomach upset,
  • blurred vision, 
  • decreased coordination, and 
  • dry mouth/nose/throat

Serious side effects of Triprolidine include:

  • mental/mood changes (such as hallucinations, irritability, nervousness), 
  • ringing in the ears,
  • trouble urinating, 
  • easy bruising/bleeding, 
  • fast/irregular heartbeat, 
  • seizures

Rare side effects of Triprolidine include:

  • none

Seek medical care or call 911 at once if you have the following serious side effects:

  • Severe headache, confusion, slurred speech, arm or leg weakness, trouble walking, loss of coordination, feeling unsteady, very stiff muscles, high fever, profuse sweating, or tremors;
  • Serious eye symptoms such as sudden vision loss, blurred vision, tunnel vision, eye pain or swelling, or seeing halos around lights;
  • Serious heart symptoms include fast, irregular, or pounding heartbeats; fluttering in the chest; shortness of breath; sudden dizziness, lightheadedness, or passing out.

What Are Dosages of Triprolidine?

Adult and pediatric dosage

Oral liquid

  • 0.5 mg/mL (Histex)
  • 0.625 mg/mL (Vanahist PD, M-Hist PD)
  • 0.938 mg/mL (Histex PD Drops)
  • 0.313 mg/mL (Vanaclear PD)

Allergic Rhinitis

Adult dosage

  • Histex: 2.5 mg (5 mL) orally every 6 hours
  • Vanahist PD or M-Hist PD: 2.5 mg (4 mL) orally every 6 hours

Pediatric dosage

Histex

  • Aged below 6 years: As directed by physician
  • Aged between 6 to 11 years: 1.25 mg (2.5 mL) orally every 6 hours or as needed
  • Aged 12 years and above : 2.5 mg (5 mL) orally every 6 hours

Vanahist PD or M-Hist PD

  • Aged below 6 years: As directed by physician
  • Aged between 6 to 11 years: 1.25 mg (2 mL) orally every 6 hours or as needed
  • Aged 12 years and above: 2.5 mg (4 mL) orally every 6 hours

Histex PD Drops

  • Aged between 4 month to 1 year: 0.313 mg (0.33 mL) orally every 4-6 hours; not to exceed 4 doses (1.33 mL) per 24 hours
  • Aged between 2 to 3  years: 0.626 mg (0.67 mL) orally every 4-6 hours, not to exceed 4 doses (2.67 mL) per 24 hours
  • Aged between 4 to 5  years: 0.938 mg (1 mL) orally every 4-6 hours; not to exceed 4 doses (4 mL) per 24 hours

Vanaclear PD

  • Aged between 4 month to 1 year: 0.313 mg (1 mL) orally every 4-6 hours; not to exceed 4 doses (4 mL) per 24 hours
  • Aged between 2 to 3  years: 0.626 mg (2 mL) orally every 4-6 hours; not to exceed 4 doses (8 mL) per 24 hours
  • Aged between 4 to 5  years: 0.938 mg (3 mL) orally every 4-6 hours; not to exceed 4 doses (12 mL) per 24 hours

What Other Drugs Interact with Triprolidine?

If your medical doctor is using this medicine to treat your pain, your doctor or pharmacist may already be aware of any possible drug interactions and may be monitoring you for them. Do not start, stop, or change the dosage of any medicine before checking with your doctor, health care provider, or pharmacist first.

  • Triprolidine has severe interactions with no other drugs.
  • Triprolidine has serious interactions with the following drugs:
    • calcium/magnesium/potassium/sodium oxybates
    • eluxadoline
    • metoclopramide intranasal

Contraindications

  • Patients taking sedatives or tranquilizers
  • Hypersensitivity; note that cross allergenicity may occur between chemically similar antihistamines

Cautions

  • May cause CNS depression, resulting in impaired physical or mental abilities; caution patients about performing tasks that require mental alertness (eg, driving, operating machinery)
  • Antihistamines may cause excitation in young children
  • Discontinue and contact physician if symptoms persist beyond 7 days or are accompanied by fever
  • Discontinue and contact physician if nervousness, dizziness, or sleeplessness occur
  • Do not exceed recommended dose
  • Discontinue therapy and ask doctor if new symptoms occur
  • Ask a healthcare professional before self-medication if experiencing breathing problems such as emphysema or chronic bronchitis, glaucoma, enlarged prostate (difficulty urinating)

Pregnancy & Lactation

  • Limited data exist
  • Generally, antihistamines are considered low risk during pregnancy; however, exposure near birth of premature infants was associated with increased risk of retrolental fibroplasia
  • Lactation
    • Distributed in human breast milk; small, occasional doses are not expected to cause adverse effects in infant
    • Larger doses may cause irritability or colicky symptoms in infant or decrease milk production in mother; nonsedating antihistamines are preferred during breastfeeding
    • Consider the developmental and health benefits of breastfeeding along with the mother’s clinical need for the drug, and any potential adverse effects on the breastfed infant from the drug or from the underlying maternal condition
